Conclusions
• BREAK-3 showed a 70% decrease in the risk of progression or
death compared with DTIC in BRAF-mutated melanoma
• Dabrafenib induced significant anti-tumor response in 53%
(INV) and 50% (IRC) of patients, respectively
• Few serious AEs and treatment-related discontinuations (3%)
with dabrafenib and DTIC
• SCC/KA were seen in 7% and photosensitivity in 3% of
dabrafenib-treated patients
• Next steps: BRAF/MEKi (dabrafenib/trametinib) combination
studies for stage IV and in the adjuvant setting

Conclusions
• The clinical activity for dabrafenib (150mg BID) in combination with
trametinib (2mg QD) is consistently superior in BRAFi-naive pts vs. BRAFi-
resistant pts
• Dual MAPK blockade may overcome primary resistance and delay
secondary resistance to BRAF inhibition
• Once BRAFi resistance occurs, the combination has limited activity with
infrequent clinical responses
– Patients with early progression on monotherapy derived minimal benefit with subsequent
combination therapy
• It is possible that early evaluation of effects of BRAF and MEK inhibitors on
tumors may provide an ability to modify treatment prior to clinical resistance

GDC-0973 in combination with vemurafenib can be delivered
safely at the respective single-agent MTDs:
vemurafenib 960 mg BID, and
GDC-0973 60 mg QD 21 days on / 7 days off
Adverse events were tolerable and manageable
Preliminary anti-tumor activity in vemurafenib-naïve patients is
encouraging
