T. melanosporum vs. T. indicum

INTRODUCTION AND BACKGROUND
In Spain the use of truffles in natural forests, has traditionally been an
important source of income in rural areas, raising in recent years a great
interest in the establishment of artificial truffle from plant micorrizada with
Tuber melanosporum Vitt., especially as a result of the new policy of the
CAP, which established important subsidies for reforestation activities.
Notwithstanding the foregoing, in Spain it is still of an official methodology
that allows to certify the quality of the mycorrhizal fungi of the genus
Tuber plants spp...
The assessment of the level of mycorrhization produced artificially by
inoculation of different plants with the fungus Tuber spp., is done currently
by different methods, depending on the country concerned, unless it exists
to date within the European Union, a norm that regulates the certification of
this type of biological material.
This work includes protocols and basic criteria for papers published for
such certification (Fisher & hills 1998, Palazón et al. 1997, Reyna 2002,
Bencivenga et al. 1996) based on morphological characters of mycorrhizal
fungi.
The following method of assessment ensures the validity of the plant
micorrizada for use in truficultura, but not the production in field of truffles.
METHODOLOGY
Control of inoculum
There are different types of esporales Inoculants and miceliares for the
production of plant micorrizada with Tuber spp. In our country today and at
the commercial level, only working with esporales inocula.
Phenomena of convergence in the morphology of mycorrhizal fungi that
make difficult or impossible the differentiation of certain species of Tuber
exist spp., including:
T. melanosporum vs. T. indicum
T. borchii vs. T. magnatum
T. brumale vs. T. brumale var. moschatum
T. aestivum vs. T. uncinatum
T. borchii vs. T. magnatum
T. brumale vs. T. brumale var.. moschatum
T. aestivum vs. T. uncinatum
In other species (T. panniferum...) the cistidios or espinulas are not present.
A previous microscopic analysis of material (fruiting bodies) that will take
place the inoculum is therefore necessary.
The method is to be expeditious and economically viable, later counting
one of the apexes micorrizados is done at magnifying glass, where it is not
feasible to differentiate species of Tuber level. A preliminary analysis of the
inoculum, practically guarantees us that Mycorrhizae which we are
expecting are inoculated Tuber species.
It will be provided nursery test tubes so that they collect samples of
serfdom (spores) that they be used to inoculate the lots. Each tube shall be
identified scoring the fungal species and number of corresponding truffle,
saving the truffles separately until its determination. The samples shall be
deposited in the center of official certification, where is added preservative
FAA until the microscopic observation of the ascospores.
Also the nursery must certify the Andalusian origin of used truffles.
Homogeneous batch identification
Shall be considered a homogeneous batch when all plants of the same are
identical:
-origin of the Acorn, with identified source coming from Andalusia.
-planting date.
-substrate cultivation.
-type of inoculum, with identified source coming from Andalusia.
-process, method and date of inoculation (in an interval of 10 days).
-irrigation regime.
Sampling
The commercial quality of the plants will be the first factor to consider,
what items will be formed, as a minimum, by 95% of plants that meet the
required criteria (Appendix 1, Peñuelas, 1993) for this purpose. This is very
important because the plants that do not comply with them, be excluded
count apices micorrizados which will be held later.
The sample size is conditioned by the high price of the plants and the
industriousness of the analysis carried out on them. There is a non-
destructive (Reyna 2000) certification methodology, with the added
advantage of taking into account the
Q. faginea, Q. robur, Q,cerris, Corylus avellana, Ostrya carpinifolia, Tilia cordata, Populus
alba, Salix caprea, Pinus pinea).
total number of Mycorrhizae for plant; It is faster and therefore economical. This method has been tested for encina, coinciding the results with other percentage protocols (Fisher & hills 1996).
Therefore volumetric Reyna (2000) protocol is used preferably for certification of holm oaks
(Quercus ilex), while, although also valid for encina, a percentage protocol described later for
other guests used to a lesser extent in truficultura will be followed (Quercus pubescens, q. faginea,
q. robur, Q, cerris, Corylus avellana, Ostrya carpinifolia, Tilia cordata, Populus alba, Salix
caprea)(, Pinus pinea).
In certain periods of the year or due to irrigation conditions, characteristic of each species of
Tuber the cistidios and/or espinulas are not present or may disappear. For this reason the analysis
of mycorrhization status shall be carried out preferably in autumn, and can also be performed in
spring. It must be taken into account that last 4-5 months from inoculation.
Identification of Mycorrhizae of Tuber spp. can be carried out according to the keys and
descriptions, among others, Meotto et to the. (1995), Zambonelli et to the. (1993) and Etayo and
Miguel's (1998), Verlhac et to the. (1990), Granetti (1995).
It is essential that samples of the plants be developed by control agent and be totally random to
avoid any type of deviation and satisfy the requirements of the statistical methods used. Plants, or
substrate samples, must be transported to the laboratory of certification refrigerated and stored at
4 ° C until they are examined, preferably within a period of 2 weeks from sampling in the nursery.
The sample size is estimated at 5 p.1000 minimum, although it is recommended that, in the early
years of certification begins with an 8 p.1000.
The number of plants to be sampled per lot, are never less than 8, independent of the size of the
lot.
1. Volumetric method Reyna (2000) for oak
1.1 Takes samples
The plants selected at random are labeled and numbered if necessary to perform any subsequent
check in them.
Sampling in volume of the root balls is performed in the following way: from the root ball of each
plant cultivated in container, two cylindrical samples of 1.4 cm in diameter are extracted with a
punch in the middle of the container and a length equal to the width to that height, which varies
with the type of container. The
contenedor.
second sample is rotated 90 ° container and removed just above or below the previous one, by removing two cylinders in a cross.
Each extracted cylinder assumes a sampled volume in the order of 7 to 9 cc., which equals
approximately 2% of the volume of the container. The cylindrical sample is extracted in a
horizontal direction. This removed the plant from the container and is placed in another which has
been already a perforation at medium altitude, and is introduced the punch printing a steady
rotation to ensure the cutting of the roots and prevent its desgajamiento or breakage. Samples
drawn in the middle of the container are considered sufficient, since no significant differences
between the upper and the middle stratum, or between the middle and the bottom, for a sample of
10 plants were obtained in previous tests.
1.2 Washing of the samples of oak
For certification discusses one of the two cylinders from each plant. It is to enter the portion of the
root ball in a plastic container and weighed in the laboratory with precision of 0.1 grams. About
100 cc are incorporated. of water to each sample next to a commercial detergent (Calgon). The
samples are subjected to sonication for 15 minutes. A new ultrasonic bath is given within 24 hours
and passed the contents of the container into a beaker of 0,5 l. incorporating water until
completing the 500 cc. Several successive incorporating water and decanted within 1 minute
decantations are performed. This is achieved the Elimination of clays, silts, and part of the organic
matter. Manipulation is done with utmost care to avoid loss of root apices.
The product is collected on a sieve with 1 mm of light and becomes washed by immersion slow in
a beaker. Product opted in the bottom of the vessel is observed again in order to check that apexes
micorrizados have not lost.
1.3 Apices count
Product of the sieve is observed to the binocular Loupe, counting all mycorrhizal inoculated
truffle, apices without micorrizar and polluting Mycorrhizae.
All references are calculated by volume, the alternative refer to sample weight gave greater
differences due to different humidity, gaps or lack of uniformity in the substrate.
With this counting we get the percentage of mycorrhization with respect to the total number of
counted apexes and the total number of mycorrhizal if we extrapolated to the total volume of the
container.
percentiles 25% (516 micorrizas/planta), 50% (1512 micorrizas/planta), 75% (2479
1.4 Criteria of validity for a plant micorrizada
He is considered by different authors (Bencivenga et 1995, Fischer & hills 1997, Palazón et al.,
1997) that from 25% of mycorrhization, according to some, and up to a 33% of mycorrhization,
according to others, the plant already is quality. Since these data are obtained, always on a
minimum of 250 apices observation, although in some cases this is not indicated, therefore this type
of criterion only guaranteed between 62 and 82 Mycorrhizae per plant.
We will consider this figure, rounded up to 100, as minimum level to qualify a plant like
micorrizada, a lower number is considered insufficient.
Altogether 7 batches studied by Reyna (2000) by sampling volume the 10th percentile stands at 260
Mycorrhizae of T. melanosporum per plant and will consider this value as the admissible minimum
so that the plant has a minimum acceptable quality. 25% Percentiles (516 Mycorrhizae per plant),
are used to delimit the rest of quality levels 50% (1512 Mycorrhizae per plant), 75% (2479
mycorrhizal plant) and 85% (2892 mycorrhizal plant), rounded, would the categories shown in the
following table:
Grades of quality of plant micorrizada with Tuber spp
number of Mycorrhizae for plant quality of mycorrhization
1-100 Insufficient
101-250 Sparse
251-500 OK
501-1500 Buena
1500-3000 Very good
> 3000 Excellent
The presence of any other Tuber, different from the inoculated species will involve the immediate
rejection of the batch. This fact should be anecdotal if the prior analysis of truffles used for the
preparation of the inoculum.
The maximum percentage of contaminated apices, must not exceed 30%. The difference between %
of micorrizados with the desired truffle and apices apexes contaminated % must be greater than or
equal to 20.
If observation of substrate samples is one without Mycorrhizae, owing that to validate a batch is
accepted only 5% of plants without any Mycorrhizae of inoculated truffle, before rejecting the
batch, will analyze it replica extracted from the same plant. In the case nor observed no mycorrhizal
fungi, are prompted to the nursery the
entero si existen o no micorrizas.
2. Percentage method for oak, Hazel, oak and other guests
2.1 Washing plants
The plants should be carefully extracted from the container with the entire root ball and dipped in a
bath of water to soak the substrate. Then, will be to remove the substrate of the roots of plants, one
by one, in a pan of water with agitations soft to not break the apices changing the water frequently
and without applying direct pressure of a stream of water. The remains of substrate that remain
attached to the roots can be removed with tweezers under a stereoscopic microscope.
2.2 First observation to the magnifying glass
Once washed the floor, horizontally immersed in a container with water. The container can be
transparent or some color at the base, which contrast with the coloration of the root, if this
facilitates the observation.
To be considered in this first observation details are as follows:
-presence or absence of Mycorrhizae
-amount of the same
-place of concentration (in the neck of the root, middle sector or apical sector) or homogeneous
distribution.
-microscope, the presence, development and overall appearance of the Mycorrhizae of inoculated
Tuber, and the presence of contaminants. At this stage the analyst must examine the apices whose
state micorricico is uncertain to facilitate subsequent count optical microscope.
Another problem affecting the quality of the plant is the absence or scarcity of trophic roots which
can lead to unreliable the percentage values of mycorrhization, the observer should take into
account and consequently reject this plant of the sample.
2.3 Count of Mycorrhizae
Immediately after the first observation proceed to remove the plant from its container, placing it on
the table, next to a rule or template that allows us to divide the root device of the plant in three
sectors (S1 = top, S2 = medium and S3 = apical), approximately the same length.
Different root fragments, belonging to 3 sectors are cut in Petri plates with water.
Each plate are taken at random several roots which have, with the help of the binocular Loupe, the
micorrizados in 50 apices apexes. According to Bencivenga et al., (1995) there is a very close
positive correlation in the percentage of mycorrhization of the first 50 numbered apices and the first
100 apices. If at the end of the counting there is a high heterogeneity in the percentages of
mycorrhization, in order to contain the error within 5%, with 95% probability, at least half of the
root tips of the plant should be analysed.
At the end you get, well, 3 percentages, which calculates the arithmetic mean, which corresponds to
the percentage of mycorrhization (PM) of the analyzed plant. All the data obtained from the
reception of the batch will be collected in physical assessment (Appendix 3) that facilitates the
control of them.
2.4 Criteria of validity for a plant micorrizada
It set minimum conditions, knowing that in some cases may be insufficient and others excessive.
This can clearly be explained if we bear in mind that the plant micorrizada, going to transplant in a
final, full of competitors biological terrain and its proportion of apices micorrizados by selected
truffle is only one factor more, within the complexity of the ecosystem in which will be to develop,
to decide in what sense is going to develop such symbiosis. A percentage of mycorrhization (PM)
of the 20 p.100, is really scarce, although it may be enough if the definitive terrain meets a number
of features, which make towards selected truffles very difficult competition. Otherwise we could
have plants with a 40 p.100 of mycorrhization, whose symbiosis will undergo a major recession due
to the excess of organisms and species competing, and may even disappear with the passage of
time. Is therefore clear that the characteristics of the plant micorrizada are very important, but it
should not lose sight the importance of other factors such as soil, its texture, its chemistry, its
microbiology, climatology, and cultural practices, which will condition the success of a truffle
plantation.
The conditions laid down have been the following:
-the minimum percentage of apices micorrizados inoculated Tuber species must be greater than or
equal to 30% (PM 30).

-the presence of any other Tuber, different from the inoculated species will involve the immediate
rejection of the batch. This fact should be anecdotal if the prior analysis of truffles used for the
preparation of the inoculum.
-the maximum percentage of contaminated apices, must not exceed 30%. The difference between %
of micorrizados with the inoculated truffle and apices apexes contaminated % must be greater than
or equal to 20.
Criteria of validity for a lot of plant micorrizada for both protocols
The homogeneous batch of plants, where at least 80% of the plants exceed the criteria of validity for
a plant and where only 5% of the plants are deprived of Mycorrhizae of inoculated truffle will be
valid for truficultura. There should be no plant with mycorrhizal fungi in a sort of different to the
inoculated Tuber.
Period of validity of certification for both protocols
If the batch does not exceed the control, the plant which constitutes it can be grown in the nursery
for a second year. If the second year the batch continues being invalid, any plant which constitutes
it may be certified as suitable for the truficultura.
It is estimated a period of validity of the certification of 6 months after the analysis of
mycorrhization. Given that the certification be carried out preferably in autumn, the distribution and
marketing of the plant must be carried in the winter or next spring.
Mycorrhization status can be modified with the permanence of the plant in the nursery, in negative
sense, by the entry of foreign fungi. If the plant is not marketed within this period, you must pass a
new control.
